The film recreates a mutiny on a stormy night off the coast of Cuba in 1839 when 53 Africans arrested in the cramped cargo holds of the Spanish slave ship La Amistad break free of their shackles. Under the leadership of Cinque, they arm themselves, take control of the ship and reclaim their freedom.
